BaroFold has developed and patented a protein processing technology that employs hydrostatic pressure to disaggregate and refold recombinant proteins from any step in a manufacturing process. The PreEMT™ process refolds proteins at higher concentrations (e.g., > 20 g/L) and shorter times (hrs), resulting in higher yields of native protein compared to conventional methods. .jpg)
With BaroFold's technology, aggregated protein at any stage in the manufacturing process is exposed to elevated pressure dissolving aggregates within minutes. Biologically active, native protein is recovered within hours at yields up to 100%, even at high protein concentrations.
The process has been tested successfully on more than 200 proteins, including:
• Mammalian cell culture derived recombinant human placental bikunin (yield increased from 55% to >95%)
• Inclusion-body-derived recombinant interferon-γ (yield ~100%)
• Recombinant human growth hormone (20 g/L ; yield ~100% )
• Multiple humanized monoclonal antibodies to recover aggregates formed during culture as well removing soluble aggregates in final product formulations
• A series of 22 “discovery-stage” proteins expressed as inclusion bodies in E. coli, which were disaggregated and refolded
